ESR study on the scavenging effects of the main ingredients in Rheum palmatum L. on superoxide anion radical Major: Prescription of TCM Postgraduate: Luo Zhiyi Supervisor: Prof. Bao Guorong ABSTRACTObjective To study the scavenging effects of Rhein、Aloe-emodin、Emodin、Physcion and Chrysophanol which were the main ingredients in Rheum palmatum L.on superoxide anion radical and to investigate the mechanism of Rheum palmatum L.in retarding aging at molecular biology lever. Method 1.Rhein、Aloe-emodin、Emodin、Physcion and Chrysophanol in Rheum palmatum L.were isolated respectively by pH gradient extraction method.And column chromatography was used to isolate and purify Physcion and Chrysophanol. 2.The melting point method、thin layer chromatography 、ultraviolet spectrophotometry and high performance liquid chromatography methods were used to identify and evaluate the purity of the the main ingredients in Rheum palmatum L. 3.The superoxide anion radical(O2˙) was produced in the alkaline dimethyl surfoxide(DMSO) system and electron spin resonance(ESR)was used to measure the signal intensity of it.The formula was:E=(h0-hx)/h0×100%(In it E was the clearance rate, h0 was the signal intensity of O2˙before the drug was added,hx was the signal intensity of O2˙after the drug was added).The method was used to observe the scavenging effect on the superoxide anion radical(O2˙)of Aloe-emodin、Rhein、Emodin、Physcion and Chrysophanol through comparing the signal intensity of O2˙which the drugs had been added with those not added ones,VitC was the positive control ones. Results All of the Rhein、Aloe-emodin、Emodin、Physcion and Chrysophanol were effective in scavenging the superoxide anion radical(O2˙)produced in the alkaline DMSO system,and the ability was dose- dependent.When the clearance rate reached 100%,the concentrations of Rhein、Chrysophanol、Emodin、Aloe-emodin、 Physcion、and VitC were 0.06mg/ml、0.39mg/ml、0.46mg/ml、6.82mg/ml、1.18mg/ml、0.72mg/ml. Conclusions:Rheum palmatum L.has the effect of retarding aging.Perhaps this is relevant to the scavenging effect on the superoxide anion radical(O2˙)of the main ingredients in the herb.
